Kisspeptin-10 peptide

Kisspeptin-10 is a short peptide used in laboratory and preclinical research to study regulation of the reproductive hormone axis, including GnRH and LH release. Common applications include investigation of puberty timing, fertility mechanisms, metabolic influences on reproduction, and kisspeptin receptor signaling. While clinical uses remain under study, research has potential to inform diagnostics and therapies for reproductive endocrine disorders.

What Is Kisspeptin-10?

Kisspeptin-10 (KP-10) is a synthetic decapeptide corresponding to the active C-terminal region of the KISS1 gene product. Its sequence is YNWNSFGLRF-NH₂, including an amidated terminal phenylalanine.

This bioactive member of the kisspeptin family activates KISS1R, also called GPR54, a G-protein-coupled receptor involved in reproductive signalling. The receptor and its ligand are distinct molecules and should be identified correctly when selecting experimental materials.

Characteristic Detail
Sequence YNWNSFGLRF-NH₂
Molecular formula C₆₃H₈₃N₁₇O₁₄
Molecular weight Approximately 1302.5 g/mol
Stated pack quantity 10mg

Scientific Background

The metastasis suppressor gene KiSS-1 encodes a precursor from which several related peptides are produced. Work by Kotani and colleagues helped identify these molecules as natural ligands for GPR54, connecting earlier melanoma research with receptor pharmacology.

Evidence about the precursor, longer peptides and KP-10 should be distinguished carefully. A result obtained with one KISS1-derived preparation does not automatically apply to every related sequence or commercial product.

Research Applications

Kisspeptin-10’s role in the hypothalamic-pituitary-gonadal axis makes it relevant to studies of gonadotropin-releasing hormone signalling. Investigations of this pathway have contributed to understanding reproductive development and puberty. The HPG axis provides a biological framework, but an isolated-cell experiment examines only selected parts of that system.

Application Experimental consideration
Receptor activation Distinguish binding from downstream responses
Neuroendocrine signalling Select endpoints appropriate to the model
Structure–function comparisons Account for sequence and terminal modifications
In-vitro assays Include suitable controls and concentration ranges

Researchers may transfect CHO cells with a receptor construct to investigate signalling in a defined system. Expression levels and cell background can influence the results, so these conditions should be recorded alongside exposure time and assay methods.

Cell Migration and Tumour-Related Studies

Separate experiments have examined KP-10 as an inhibitor of angiogenesis-associated processes. Published work reported effects on endothelial cell migration, invasion and tube formation under specified conditions. These findings support further investigation rather than establishing an anticancer treatment.

Use in research should distinguish cell movement from changes in survival or proliferation. For example, a Cell Counting Kit-8 assay provides a metabolic viability readout; it does not directly measure migration. Complementary methods help researchers interpret whether an observed response reflects the mechanism being investigated.

Quality and Analytical Documentation

When selecting high-purity material, request the batch-specific certificate of analysis. Review the reported purity together with identity evidence, such as mass spectrometry, and check that the records match the supplied product.

Purity alone does not establish peptide quantity, sterility or biological activity. Preparation conditions and appropriate controls remain essential in research settings.

Preparation and Storage

The product is supplied as a lyophilised powder. Follow its documented solvent and handling instructions rather than assuming that one preparation method suits every assay.

If the supplier specifies that the material should be stored at -20°C, maintain those conditions and protect it from unsuitable light and moisture exposure. Where instructed, allow the sealed container to reach room temperature before opening to limit condensation.

Prepared solutions require their own validated storage limits. Use suitable aliquots to avoid repeated freeze-thaw cycles where the protocol recommends this. A clear solution does not confirm purity, sterility or successful preservation of activity.

The supplied reagent is not intended for use in humans or animals. Its research designation should not be presented as evidence of MHRA approval or suitability for clinical treatment.
